llknown formulas, which has been traditionally used as a tonic prescription nourishingyin of kidney in China for hundreds of years. Recently, clinical observations showed that it could alleviate menopausal symptoms, and the therapeutical effect was better than estrogen used alone. Considering its little side effect, Zuo Gui Yin Decoction is now widely used as a safer way to treat menopausal symptoms in clinic, but the knowledge of its action mechanism is limited. Since the basic reason of menopausal symptoms is reduced estrogen level due to ovarian aging, the present study was designed to investigate the effect and molecular mechanism of Zuo Gui Yin Decoction on ovarian function of rats during perimenopause. This will provide more theoretical support for the application of Zuo Gui Yin Decoction in clinic, and contribute to the exploration and utilization of traditional Chinese medicine. In this study, we mainly focused on its’ effects on the expression of several genes related to ovarian estradiol production, ovarian cell apoptosis pathway and ovarian local factors regulating microcirculation. The animal model of perimenopausal rats was established by unforced aging. Perimenopausal rats were treated by intragastric administration (ig) with low ( g/kg), middle ( g/kg) and high (31 g/kg) dose of Zuo Gui Yin Decoction for 8 weeks.
